چکیده
Anti-microbial peptides (AMPs) are important factors of the innate immune system which perform a comprehensive spectrum biological activities. The present study aims to examine expression, purification, and cytotoxic effects recombinant PI–laterosporulin10 (LS10) defensing-like peptide. PI is transmembrane peptide that helps increase penetration specificity targeting. PI–LS10 sequence was optimized fused Xa protease site achieve higher levels expression. Then Xa–PI–LS10 cloned into pET-32a (+) vector expressed in Escherichia coli BL21 (DE3). fusion protein with molecular weight about 25.3 kDa analyzed using SDS-PAGE. maximum expression observed 8 h post-induction. After enzymatic cleavage, purity > 300 µg/ ml as final yield. showed anti-cancer characteristics against breast cancer cell lines. MCF7 (non-triple negative) MDA-MB-231 (triple cells were treated different concentrations order evaluate their activity efficiency targeting triple negative cells. Our results suggest had significant effect comparison normal line. Triple (MDA-MB-231) proliferation inhibited 72 after treatment compare In conclusion, produced successfully its favorable line confirmed.

Design of a Novel Recombinant Multi-Epitope Vaccine against Triple-Negative Breast Cancer
Background: Triple-negative breast cancer (TNBC) is determined by the absence of ERBB2, estrogen and progesterone receptors’ expression. Cancer vaccines, as the novel immunotherapy strategies, have emerged as promising tools for treating the advanced stage of TNBC. The aim of this study was to evaluate Carcinoembryonic antigen (CEA), Metadherin (MTDH), and Mucin 1 (MUC-1) proteins as vaccine ...
متن کاملCytotoxicity and anti-tumor effects of new ruthenium complexes on triple negative breast cancer cells
Triple-negative breast cancer (TNBC) is a highly aggressive breast cancer subtype. The high rate of metastasis associated to the fact that these cells frequently display multidrug resistance, make the treatment of metastatic disease difficult. Development of antitumor metal-based drugs was started with the discovery of cisplatin, however, the severe side effects represent a limitation for its c...
متن کاملFuture of Triple Negative Breast Cancer: Can Immunotherapy Treat This Deadly Subtype of Breast Cancer?
Triple negative breast cancer (TNBC): challenges and solutions via the immune cells TNBC is one of the most complicated types of breast cancer to treat. It is generally diagnosed based on the absence of three receptors: estrogen, progesterone, and human epidermal growth factor receptor 2 (HER2) and is thus defined as a triple negative. TNBC is often more aggressive with lower survival rates...
متن کاملClinicopathological Features of Non-metastatic Triple Negative Breast Cancer
Background: Triple negative breast cancer (TNBC) is reported to be associated with a high risk of recurrence, poor overall survival (OS), and disease-free survival (DFS) rates. This study evaluated the clincopathological features and survival of non-metastatic TNBC women in the capital of Iran compared with other areas of the world. Methods: In a retrospective study, 119 women with TNBC based ...
متن کاملAssociation of HDAC8 Expression with Pathological Findings in Triple Negative and Non-Triple Negative Breast Cancer: Implications for Diagnosis
Background: Previous data have shown the tumorigenicity roles of HDAC8 in breast cancer. More recently, the oncogenic effects of this molecule have been revealed in TNBC. The present study aimed to determine the diagnostic value of HDAC8 for the differentiation of TNBC from nTNBC tumors. Methods: A total of 50 cancerous and normal adjacent tumor specimens were obtained, and the clinical and pat...
